How to actually do it
- Draw the diluent into a syringe. A larger syringe than the dosing one is easier here.
- Aim down the inside wall of the vial, not at the powder. A jet straight onto the powder is the most common way to damage a peptide during reconstitution.
- Let it run in slowly under its own vacuum rather than forcing it.
- Swirl gently until dissolved. Do not shake. Shaking foams the solution and shears the peptide; foam also makes an accurate draw impossible.
- If anything remains undissolved after a few minutes, leave it and swirl again rather than agitating harder.
- Label the vial with the date of first puncture — that is what the beyond-use date counts from, not the date you bought it.
These are handling conventions, not a claim about any specific compound's stability. Where a compound has its own handling requirement on file, it is on that compound's record.
The formula
mass ÷ diluent = concentration dose ÷ concentration = volume volume × factor = units on the barrel (U-100 → 100, U-40 → 40, U-500 → 500)
A U-40 syringe multiplies by 40 instead of 100, and a U-500 by 500. Getting that factor wrong is the most consequential arithmetic error in this domain, which is why the syringe is a field rather than an assumption.

Working backwards
The most useful direction, and the one most calculators lack. If you want a 250 mcg dose to land on exactly 10 units — easy to draw, hard to misread — this is the diluent that gets you there:
10 units ÷ 100 = 0.1 mL per dose 250 mcg ÷ 0.1 mL = 2500 mcg/mL needed 5 mg ÷ 2.5 mg/mL = 2 mL of diluent
Round numbers on the barrel are worth engineering for. A dose that reads as "10 units" is misread far less often than one that reads as "7.5".
